Inoculated means that some bacteria have been put into the tube and allowed to grow. The stuff in the tube is agar (sort of like Jello) mixed with some food the bacteria like to eat. We only put the bacteria along a line in the agar (we stab a wire with some bacteria on it into the agar). This makes it easy to see if they grow. It also means the bacteria do not have any air to breath. We get our energy by breathing air and using that to burn up our food. Without air (oxygen actually) we would quickly die. These bacteria can live without air if you give them light (otherwise they die just like we do). They use the light to make energy instead of burning the food with oxygen.
